Overview

αCD19 CAR-Tconv cells are conventional CD4+ and CD8+ T cells (Tconv) genetically modified to express a chimeric antigen receptor (CAR) specific for the CD19 antigen. These cells typically utilize a second-generation CAR construct containing a 4-1BB costimulatory domain. In neuro-oncology research, such as studies conducted at the University of Calgary, αCD19 CAR-Tconv cells are frequently employed as a negative control to evaluate the efficacy and safety of novel CAR-T platforms (e.g., CAR-DNT cells) targeting glioblastoma-associated antigens like CD276 (B7-H3). While CD19-targeted CAR-T therapies are clinically established for B-cell malignancies, in the context of solid tumor research, they serve as a benchmark for assessing CAR-mediated versus CAR-independent cytotoxicity and the risk of xenogeneic graft-versus-host disease (GvHD).
