αCD276 CAR-Tconv cells are an experimental chimeric antigen receptor (CAR) T-cell therapy targeting CD276 (also known as B7-H3), a transmembrane protein overexpressed in various solid tumors, including glioblastoma (GBM). These cells are derived from conventional CD4+ and CD8+ T cells (Tconv) and engineered with a second-generation CAR construct containing a 4-1BB costimulatory domain. Developed by researchers at the University of Calgary, αCD276 CAR-Tconv cells have demonstrated potent anti-tumor activity in preclinical orthotopic xenograft models of GBM, effectively targeting both the tumor core and the infiltrative edge. However, their use in allogeneic settings is limited by the induction of severe xenogeneic graft-versus-host disease (GvHD), leading to the investigation of alternative cell platforms such as double-negative T (DNT) cells.
