Overview

αCD33 CAR-T is a chimeric antigen receptor (CAR) T-cell therapy designed to target the CD33 antigen, a transmembrane protein frequently expressed on the blasts of patients with acute myeloid leukemia (AML). The specific construct described in preclinical research by the Hospital Clinic de Barcelona and the Josep Carreras Leukaemia Research Institute is a second-generation CAR comprising an anti-CD33 single-chain variable fragment (scFv), a CD28 costimulatory domain, and a CD3ζ signaling domain. This therapeutic approach aims to induce T-cell mediated lysis of CD33-positive leukemic cells. While evaluated as a monotherapy in preclinical models, it is also being integrated into dual-targeting strategies (e.g., CD33/CD123) to mitigate antigen escape and improve clinical outcomes in relapsed or refractory AML.
