**αCD40 immunosome** is an academic, preclinical liposomal immunosome developed for glioblastoma immunotherapy. The formulation presents an agonistic anti-CD40 antibody on the liposome surface and encapsulates the small-molecule immunomodulator RRX-001. It is designed to improve tumor retention and mitigate the systemic immune and hepatic toxicities associated with free αCD40 while remodeling the glioblastoma tumor microenvironment toward antitumor immunity. In murine GL-261 glioblastoma models, the formulation increased intratumoral cytotoxic T cells, B cells, dendritic cells, and activated macrophages; reduced regulatory T cells, M2 macrophages, myeloid-derived suppressor cells, and PD-L1 expression; and produced tumor-free survival in treated mice.
