**αCD45-cs21** is a preclinical, CD45-targeted immunocytokine comprising an anti-CD45 immunoglobulin conjugated to cs21, a computationally stabilized murine interleukin-21 variant. CD45 binding localizes the cytokine to leukocytes following intratumoral administration, while cs21 activates interleukin-21 receptor signaling and downstream STAT3 phosphorylation in immune cells. In syngeneic B16F10 melanoma models, intratumoral αCD45-cs21 increased pSTAT3 in CD45-positive cells in tumor-draining lymph nodes and improved median survival as monotherapy; combinations with IL-12 or IL-15 showed preclinical synergy without apparent toxicity.
