**αCD45-IL12** is an investigational immunocytokine fusion protein consisting of an anti-CD45 antibody linked to single-chain interleukin-12 (IL-12p70). It targets the ubiquitous leukocyte receptor **CD45** to enable localized retention on immune cells, enhancing cytokine signaling duration and concentration at the tumor site while minimizing systemic toxicity. Developed as part of a preclinical cytokine therapy platform, it promotes tumor inflammation, antigen release by tumor-infiltrating leukocytes, and reprogramming of tumor-specific CD8+ T cells in tumor-draining lymph nodes to an antiviral-like effector state, leading to eradication of treated tumors and abscopal effects on distal lesions in syngeneic mouse models of melanoma (B16F10), colon carcinoma (MC38), and others when sequenced with αCD45-IL-15.
