Overview

αGAL-9Ab is an experimental monoclonal antibody targeting Galectin-9 (GAL-9), currently under investigation for the treatment of various acute leukemias, including acute lymphoblastic leukemia (ALL) and acute myeloid leukemia (AML). Developed by researchers at Emory University, the antibody exhibits direct cytotoxicity against B-ALL, T-ALL, and AML cells. Mechanistically, αGAL-9Ab treatment leads to the deregulation of checkpoint kinase 1 (CHEK1) and the anti-apoptotic protein myeloid leukemia 1 (MCL-1). Pre-clinical studies have demonstrated that αGAL-9Ab synergizes with CHEK1 inhibitors (such as GDC-0575, prexasertib, and PF477736) and shows superior efficacy compared to standard chemotherapy in xenograft models of T-ALL. Pediatric leukemia cell lines appear particularly sensitive to this treatment approach.
