αM.CAR4 T cells (also known as anti-MUC1-CAR4 T cells) are fourth-generation chimeric antigen receptor (CAR) T cells designed to target Mucin 1 (MUC1), a transmembrane glycoprotein overexpressed in various solid tumors, including breast cancer and cholangiocarcinoma. Developed by researchers at Mahidol University, these CAR-T cells feature a fourth-generation CAR construct containing an anti-MUC1 single-chain variable fragment (scFv) linked to three costimulatory domains (CD28, CD137, and CD27) and the CD3ζ signaling domain. In preclinical studies, αM.CAR4 T cells demonstrated specific cytolytic activity against MUC1-positive tumor cells, enhanced proliferation, and reduced expression of the exhaustion marker PD-1 compared to earlier-generation CAR-T cells.
