The αPD-L1 diabody (αPD-L1 Db) is a research-stage immunocytokine construct designed to enhance the efficacy of immune checkpoint inhibition in poorly immunogenic tumors. Developed by researchers at the University of Pittsburgh, the construct consists of a diabody targeting programmed death-ligand 1 (PD-L1) fused to interleukin-15 (IL-15), a cytokine that acts as an effector immune cell agonist. This bifunctional molecule aims to simultaneously block the PD-1/PD-L1 inhibitory pathway and stimulate immune cells via the IL-15 receptor pathway. Preclinical studies presented at AACR 2024 demonstrated that the αPD-L1-IL-15 Db, when combined with radiation therapy, significantly improves survival and tumor growth delay in immune-resistant murine head and neck cancer models. The construct is produced via transfection into Expi-CHO cells and purified using Protein L affinity chromatography.
