The αPD-L1-IL-15 diabody (αPD-L1-IL-15 Db) is a preclinical-stage immunocytokine construct developed by researchers at the University of Pittsburgh. It is a recombinant fusion protein consisting of an anti-programmed death-ligand 1 (PD-L1) diabody linked to interleukin-15 (IL-15). This dual-action molecule is designed to simultaneously block the PD-1/PD-L1 immune checkpoint and provide localized IL-15-mediated stimulation of effector immune cells, such as T cells and NK cells, within the tumor microenvironment. Preclinical studies presented at AACR 2024 demonstrated that the αPD-L1-IL-15 diabody, particularly when combined with radiation therapy to upregulate PD-L1 expression, significantly improves tumor growth delay and overall survival in immune checkpoint inhibitor-resistant models of head and neck cancer.
