Overview

αvβ3 CAR-T cells are a type of Chimeric Antigen Receptor (CAR) T-cell therapy engineered to specifically target the αvβ3 integrin. This cell therapy is being investigated for its potential in treating breast cancer brain metastasis (BCBM). Pre-clinical studies have demonstrated that when combined with focused ultrasound-mediated blood-brain barrier opening (FUS BBBO), αvβ3 CAR-T cells significantly constrain BCBM outgrowth compared to monotherapy. The FUS BBBO appears to enhance the frequency of circulating CAR-T cells and increase the proportion of CD8+ CAR-T cells within BCBM-bearing brains, leading to elevated tumor-localized T cell infiltration.
