β-catenin fragment 529-581 mRNA-LNP is an experimental mRNA-based therapeutic designed to target leukemia stem cells (LSCs) by disrupting the AKT-Wnt/β-catenin signaling pathway. Developed by researchers at the Stowers Institute for Medical Research, this drug utilizes lipid nanoparticle (LNP) technology to deliver mRNA encoding a specific 53-amino acid fragment (residues 529-581) of the β-catenin protein. This fragment acts as a peptide inhibitor that interferes with the interaction between AKT and β-catenin, specifically targeting the AKT phosphorylation site at Ser552. By inhibiting this pathway, the treatment aims to reduce LSC self-renewal, overcome chemoradiotherapy resistance, and reprogram anti-tumor immunity in conditions such as acute myeloid leukemia (AML) and T-cell acute lymphoblastic leukemia (T-ALL).
