Drug intelligence / Profile preview
ΔV1 HIV envelope immunogen is an investigational HIV vaccine candidate designed by deleting the variable region 1 (V1) loop from the HIV envelope glycoprotein (gp120 or gp160, specifically based on the clade A/E A244 strain). The V1 loop acts as an immunodominant decoy that typically distracts the immune system from targeting more conserved regions. By removing the V1 loop, the immunogen redirects the antibody response toward the conserved, protective α-helical V2 loop, enhancing antibody-dependent cellular cytotoxicity (ADCC) and promoting anti-inflammatory mucosal immunity (such as CD14+ monocyte-mediated efferocytosis and IL-17+ NKp44+ innate lymphoid cell activation). This vaccine candidate is being advanced to clinical evaluation in the Phase I CLEAR (Combined Long-Term Efferocytosis and ADCC Responses) clinical trial, where it will be tested in combination with Alum and ALFQA (Army Liposome Formulation + QS21) adjuvants.
