MAPP (D-erythro-2-(N-myristoylamino)-1-phenyl-1-propanol) is a potent and selective small molecule inhibitor of alkaline ceramidase. By inhibiting the conversion of ceramide to sphingosine, MAPP effectively increases intracellular levels of ceramide, a key pro-apoptotic signaling lipid. This mechanism makes MAPP a valuable research tool in the study of sphingolipid metabolism and its role in cell growth, differentiation, and apoptosis. In various cancer cell lines, MAPP-induced ceramide accumulation has been shown to inhibit cell proliferation and induce cell cycle arrest, particularly in the G0/G1 phase. While primarily used as a pharmacological probe in biochemical research, its ability to modulate sphingolipid signaling pathways has positioned it as a lead compound for investigating potential therapeutic interventions in oncology and inflammatory skin conditions.
