Overview

(2Z)-N-biphenyl-4-yl–2-cyano–3–cyclopropyl–3–hydroxyprop–2-enamide is an experimental small molecule inhibitor of dihydroorotate dehydrogenase (DHODH), the enzyme responsible for the only redox step in de novo pyrimidine biosynthesis. It was designed using structure-based drug discovery approaches to bind the ubiquinone binding site of human and Plasmodium falciparum DHODH. The compound has demonstrated inhibitory activity against these enzymes in vitro and has been structurally characterized in complex with human DHODH[10]. There are no approved indications or clinical trials reported for this compound[1][10].
