Overview

**(5Z)-7-oxozeaenol** is a resorcylic acid lactone of fungal origin, specifically isolated from *Curvularia* species, and acts as a highly potent, selective, irreversible, and ATP-competitive inhibitor of transforming growth factor-β-activated kinase 1 (TAK1, a MAPKKK). It exhibits strong anti-inflammatory and cytotoxic properties, inhibits the NF-κB signaling pathway, and blocks the production of pro-inflammatory cytokines (IL-17A, IFN-γ, TNF-α, IL-6). It also inhibits other kinases such as VEGF receptor 2 (VEGFR2) and PDGFRβ, but with less potency. (5Z)-7-oxozeaenol has demonstrated efficacy in animal models of CNS autoimmune diseases (such as experimental autoimmune encephalomyelitis, EAE—a model of multiple sclerosis), alleviating symptoms by reducing CNS inflammation, demyelination, and microglial activation[1][3][4][5][6].
