Overview

(pGlu-Gln)-CCK-8 is a synthetic, N-terminally modified analogue of the endogenous peptide hormone cholecystokinin octapeptide (CCK-8). The modification involves substitution at the N terminus with pyroglutamyl-glutaminyl residues, which confers resistance to enzymatic degradation and enhances biological activity compared to native CCK-8. This peptide acts as an agonist at cholecystokinin receptors, particularly the CCK1 receptor. In preclinical studies in mouse models of obesity and diabetes, (pGlu-Gln)-CCK-8 demonstrated potent insulinotropic effects (stimulating insulin secretion), appetite suppression, weight-lowering properties, improved glucose tolerance and insulin sensitivity, and reduced lipid accumulation in tissues. These effects are mediated by activation of pancreatic β-cell function and satiety pathways via CCK receptor signaling[1][2][3]. The compound has been investigated primarily for its potential therapeutic application in metabolic diseases such as obesity and type 2 diabetes.
