Overview

(S)-iclaprim is the S-enantiomer of iclaprim, a synthetic small molecule antibiotic in the diaminopyrimidine class. It acts as a competitive inhibitor of bacterial dihydrofolate reductase (DHFR), disrupting folate synthesis and thereby inhibiting bacterial DNA replication and cell division. Like racemic iclaprim, it demonstrates potent activity against Gram-positive bacteria including methicillin-resistant Staphylococcus aureus (MRSA) and Streptococcus pneumoniae resistant to multiple antibiotics. Iclaprim has been investigated primarily for intravenous use in acute bacterial skin and skin structure infections (ABSSSI) and complicated skin and skin structure infections (cSSSI). The S-enantiomer specifically refers to the stereochemistry at its chiral center[1][2][4].
