Overview

(South)-Methanocarba-Thymidine is a synthetic nucleoside analogue structurally related to thymidine and designed as a conformationally restricted mimic of natural nucleosides. It acts primarily as an inhibitor of thymidine kinase 1 (TK1), an enzyme involved in the salvage pathway of pyrimidine synthesis and essential for DNA replication in both normal and viral-infected cells[2][7][8]. This compound has been studied experimentally for its ability to inhibit the growth of herpes simplex virus type 1 (HSV‑1) by targeting HSV‑encoded thymidine kinase; it is not approved for clinical use but serves as a discovery agent in antiviral research[7][8]. Its mechanism involves competitive inhibition at the active site of TK enzymes.
