Overview

1-Deaza-Adenosine is a synthetic small molecule and an analog of adenosine in which the nitrogen at position 1 of the purine ring is replaced by a carbon. It acts as a potent inhibitor of adenosine deaminase (ADA), with a Ki value of approximately 0.66 μM[4][5][8]. By inhibiting ADA, it prevents the deamination and subsequent breakdown of adenosine and related nucleosides. This mechanism leads to increased levels of adenosine in cells and tissues. Preclinical studies have shown that 1-Deaza-Adenosine exhibits antitumor activity in vitro against various leukemia cell lines such as HeLa, KB, P388, and L1210[5][8]. It has been investigated primarily for its potential use as a chemotherapy agent for lymphoproliferative disorders but remains experimental with no approved clinical indications[1][3].
