Overview

1-methyl-N-butynyl-tetrahydroisoquinoline is a synthetic small molecule derivative of the endogenous neuroprotectant 1-methyl-1,2,3,4-tetrahydroisoquinoline (1-MeTIQ). It has been investigated for its potential in treating Parkinson's disease, specifically for its ability to mitigate motor symptoms and stabilize dopamine transporter (DAT) levels. In preclinical mouse models using the neurotoxin MPTP, the compound significantly reduced bradykinesia and prevented the loss of DAT expression. However, it did not demonstrate the same level of neuroprotection as its N-propargyl analog, as it failed to prevent the reduction of striatal dopamine levels or the loss of tyrosine hydroxylase-positive neurons in the substantia nigra. This suggests that while the N-butynyl modification confers some symptomatic benefits, the specific structure and length of the N-substituent are critical for full neuroprotective activity.
