Overview

10-butyl ether minocycline (BEM) is a novel, non-antibiotic derivative of minocycline designed to retain the anti-inflammatory, antioxidant, anti-migratory, matrix metalloproteinase (MMP) inhibitory, and neuroprotective properties of minocycline without its antimicrobial activity. BEM exhibits a nearly complete loss of antibiotic effect against common bacteria but maintains potent inhibition of microglial activation, MMP-9 inhibition, endothelial cell migration, and reactive oxygen species (ROS) reduction. It is being investigated for potential use in neuroimmune and inflammatory disorders, notably as a treatment for alcohol use disorder (AUD), and has demonstrated efficacy in reducing alcohol consumption in animal models[6][8]. The drug has improved MMP-9 inhibition over minocycline and is not toxic to mitochondria at high doses[6].
