Overview

10074-G5 is a small-molecule research compound that functions as an inhibitor of the c-Myc-Max dimerization process. It binds to the basic helix-loop-helix-leucine zipper (bHLH-ZIP) domain of the c-Myc oncoprotein, causing a conformational distortion that prevents c-Myc from forming a functional heterodimer with its partner protein, Max. Because c-Myc must dimerize with Max to bind DNA and regulate the expression of genes involved in cell proliferation and survival, 10074-G5 effectively blocks c-Myc-driven transcriptional activity. The compound has shown in vitro efficacy in inhibiting the growth of cancer cell lines such as Daudi Burkitt's lymphoma and HL-60 leukemia cells. It is currently used as a chemical probe in oncology research and is not an approved therapeutic agent.
