Overview

101-PGC-005 is a Type IA prodrug of dexamethasone designed to selectively target CD206+ (mannose receptor-positive) activated macrophages. This targeted delivery aims to enhance the anti-inflammatory efficacy of dexamethasone while minimizing systemic side effects, particularly suppression of the hypothalamic-pituitary-adrenal (HPA) axis. The drug is being developed for use in combination with standard of care therapies for hospitalized patients with moderate COVID-19 and has also received FDA Rare Pediatric Disease Designation for systemic juvenile idiopathic arthritis (sJIA) flares. In clinical trials, 101-PGC-005 is administered as a bolus injection and compared against conventional dexamethasone therapy, with all patients receiving additional standard treatments as appropriate[1][2][3][4][5].
