Drug intelligence / Profile preview
101-PIF-GC-005 (also known as 101-PGC-005) is an investigational, macrophage-targeted, pH-sensitive Type IA prodrug of dexamethasone developed by 101 Therapeutics, PIF Partners, and Laxai Life Sciences. The drug is designed to selectively target CD206 (mannose receptor) expressed on pathogenic, overactive macrophages. Upon receptor-mediated endocytosis, the acidic endosomal environment triggers the release of active dexamethasone intracellularly, inducing apoptosis of pathogenic macrophages and dampening hyperinflammation. This targeted delivery mechanism avoids systemic glucocorticoid exposure, thereby preventing the suppression of the hypothalamic-pituitary-adrenal (HPA) axis and other systemic steroid toxicities. 101-PIF-GC-005 is in Phase 3 clinical development for the treatment of acute respiratory distress syndrome (ARDS) induced by COVID-19 and is being evaluated for systemic juvenile idiopathic arthritis (sJIA) flares, Bundibugyo Ebola virus disease, and other macrophage-driven inflammatory conditions.
