10B-bare liposomes are experimental non-PEGylated liposomal delivery vehicles containing Boron-10 (10B) compounds, designed for use in Boron Neutron Capture Therapy (BNCT). These liposomes are typically composed of phospholipids such as distearoylphosphatidylcholine (DSPC) and cholesterol but lack the polyethylene glycol (PEG) surface modification that characterizes long-circulating liposomes. In the BNCT paradigm, these liposomes are administered intravenously to accumulate 10B atoms within tumor cells. The tumor is then irradiated with thermal neutrons, which are captured by the 10B nuclei, triggering a nuclear fission reaction (10B(n,α)7Li). This reaction releases high-energy alpha particles and lithium-7 nuclei that travel short distances (5-9 micrometers), causing localized, lethal double-strand DNA breaks in the target tumor cells while minimizing damage to adjacent healthy tissue. In preclinical models, such as AsPC-1 human pancreatic carcinoma xenografts, bare liposomes are often used as comparative controls to evaluate the superior pharmacokinetic profile and tumor-suppressive efficacy of PEGylated 10B-liposomes.
