10B-PEG-liposomes are a specialized drug delivery system designed for Boron Neutron Capture Therapy (BNCT). The formulation consists of PEGylated liposomes composed of dipalmitoylphosphatidylcholine (DPPC), cholesterol, and distearoylphosphatidylethanolamine-polyethylene glycol 2000 (DSPC-PEG2000), which encapsulate a Boron-10 (10B) compound. The polyethylene glycol (PEG) coating provides "stealth" properties, enhancing the circulation time and allowing the liposomes to accumulate in tumor tissues via the enhanced permeability and retention (EPR) effect. In BNCT, the 10B atoms within the tumor cells capture thermal neutrons during irradiation, triggering a nuclear fission reaction that releases high-energy alpha particles and lithium-7 nuclei. These particles have a very short range (approximately 5-9 micrometers), resulting in highly localized destruction of tumor cells while minimizing damage to surrounding healthy tissue. Preclinical studies have demonstrated that intravenous injection of these liposomes can suppress the growth of human pancreatic carcinoma xenografts in vivo.
