10D12 is a murine monoclonal antibody directed against the CD6 co-stimulatory receptor. It serves as a surrogate for clinical-stage anti-CD6 antibodies, such as itolizumab, in preclinical research. 10D12 works by blocking the CD6-ALCAM (Activated Leukocyte Cell Adhesion Molecule) pathway, which is a key regulator of T cell activation, proliferation, and migration. In studies of immune-mediated diseases like lupus nephritis, 10D12 has been shown to reduce renal inflammation, decrease the infiltration of pathogenic T cells and macrophages, and improve clinical markers of kidney function. The antibody was developed and studied through collaborations involving Equillium, the Albert Einstein College of Medicine, and the Cleveland Clinic.
