10F04 TCR-T is an investigational T cell receptor engineered T cell therapy that utilizes autologous human T cells transduced with the 10F04 TCR, which is specifically restricted to HLA-DRA/DRB1*09:01 and recognizes the human papillomavirus type 18 (HPV18) E7 protein, peptide region 84-98. This approach, derived from a patient research cohort benefiting from multiple antigen stimulating cellular therapy, allows both CD4^+ and CD8^+ T cells to target and kill HPV18 E7-expressing tumor cells, particularly in cervical cancer. Following murinization and codon optimization for improved surface expression and functional activity, 10F04 TCR-T cells demonstrate robust and durable antitumor activity in vitro and in mouse xenograft models, with polyfunctional cytokine release and tumor lysis. The therapy is currently under investigation in a Phase I clinical trial for HPV18-positive cancers, especially metastatic cervical cancer. The modality is T cell receptor engineered T cell therapy, and the mechanism is antigen recognition via a synthetic TCR directed against HPV18 E7 protein presented on HLA class II (DRB1*09:01)[2][3].
