10H10 is a monoclonal antibody that targets human Tissue Factor (TF), also known as Coagulation Factor III. It is specifically characterized by its ability to selectively inhibit TF-mediated intracellular signaling—particularly the activation of Protease-Activated Receptor 2 (PAR2) by the TF-Factor VIIa complex—while typically sparing the procoagulant (clotting) function of TF. This functional selectivity makes 10H10 a critical tool in oncology and inflammation research, as it allows for the study of TF's role in cancer cell migration, metastasis, and the tumor microenvironment independently of its role in the coagulation cascade. A humanized version of this antibody, designated as ALT-836 (or Sunol-cH10H10), has been evaluated in clinical trials for indications including acute lung injury (ALI), acute respiratory distress syndrome (ARDS), and solid tumors, where it aims to block TF-driven inflammatory and metastatic pathways without increasing the risk of systemic bleeding.
