10Pn-PD-DiT-dPly-PhtD is an investigational next-generation combination pneumococcal vaccine developed by GSK. It is a protein-polysaccharide conjugate vaccine that combines the licensed 10-valent pneumococcal non-typeable *Haemophilus influenzae* protein D conjugate vaccine (PHiD-CV, brand name Synflorix) with two additional conserved pneumococcal protein antigens: detoxified pneumolysin (dPly) and pneumococcal histidine triad protein D (PhtD). The vaccine is designed to provide broad protection against *Streptococcus pneumoniae* by targeting 10 specific capsular polysaccharides as well as virulence proteins that are conserved across most pneumococcal strains. Additionally, the use of protein D from non-typeable *Haemophilus influenzae* (NTHi) as a carrier protein provides protection against NTHi-related infections. It has been evaluated in Phase II clinical trials for safety, immunogenicity, and impact on nasopharyngeal carriage in infants and children.
