10R-siRNA is a preclinical peptide-siRNA nanocomplex designed for the tumor-specific targeting and silencing of lactate dehydrogenase C (LDHC). It consists of a cell-penetrating 10R polyarginine peptide electrostatically complexed with an siRNA targeting LDHC. LDHC is highly expressed in certain tumors, such as triple-negative breast cancer (TNBC), where it acts as a key regulator of the DNA damage response and tumor mitotic fidelity. By delivering the siRNA into tumor cells, 10R-siRNA downregulates LDHC expression, reducing tumor cell clonogenicity and enhancing sensitivity to DNA-damaging therapeutics like olaparib.
