11β-methyl nortestosterone dodecylcarbonate (11β-MNTDC) is an experimental, orally active synthetic androgen and potential male hormonal contraceptive. It is a dodecylcarbonate ester and prodrug of 11β-methyl-19-nortestosterone (MENT), a potent androgen that is not susceptible to 5α-reduction. 11β-MNTDC acts as a dual agonist at the androgen receptor (AR) and the progesterone receptor (PR). This combined activity suppresses the hypothalamic-pituitary-gonadal axis, leading to a significant reduction in the secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which ultimately inhibits testicular testosterone production and spermatogenesis. Developed by the Population Council and the National Institute of Child Health and Human Development (NICHD), 11β-MNTDC is designed to provide androgen replacement while maintaining contraceptive efficacy, with a potentially improved safety profile regarding the prostate compared to testosterone.
