Overview

11-1F4-CAR-GFP is an experimental chimeric antigen receptor (CAR) macrophage therapy designed for the treatment of light chain (AL) amyloidosis. Developed by researchers at Columbia University, the therapy utilizes a first-generation CAR construct consisting of a single-chain variable fragment (scFv) derived from the anti-amyloid monoclonal antibody 11-1F4 (anselamimab) fused to an intracellular Fc receptor gamma (FcRγ) signaling domain and a green fluorescent protein (GFP) tag. Unlike traditional treatments that target plasma cells to suppress the synthesis of new amyloidogenic light chains, this CAR-macrophage approach aims to actively clear existing organ-damaging amyloid fibril deposits by enhancing the phagocytic activity of macrophages. Preclinical studies have demonstrated that 11-1F4-CAR-GFP macrophages, derived from CD34+ hematopoietic stem cells, successfully target and internalize AL amyloid fibrils both in vitro and in humanized mouse models.
