11-valent pneumococcal conjugate vaccine

The 11-valent pneumococcal conjugate vaccine (11PCV) is an investigational vaccine developed by GSK for the prevention of pneumococcal disease, including invasive infections and acute otitis media. It targets 11 serotypes of *Streptococcus pneumoniae*: 1, 3, 4, 5, 6B, 7F, 9V, 14, 18C, 19F, and 23F. The vaccine utilizes a unique conjugation strategy where most serotypes are conjugated to Protein D (a surface protein of non-typeable *Haemophilus influenzae*), while others are conjugated to tetanus and diphtheria toxoids. This formulation was designed to provide broader protection than earlier vaccines and specifically to address otitis media through the Protein D carrier. Although it demonstrated efficacy in clinical trials (such as the POET study), GSK ultimately transitioned its development efforts to the 10-valent formulation, which was commercialized as Synflorix.
