This is an autologous dendritic cell (DC) vaccine therapy radiolabeled with Indium-111 (111In) for use in the treatment of glioblastoma. The vaccine is prepared by electroporating a patient's own dendritic cells with mRNA encoding the human cytomegalovirus (CMV) pp65 protein, which is fused to a lysosomal-associated membrane protein (LAMP) to enhance antigen processing and presentation. The Indium-111 label serves as a diagnostic tracer, allowing clinicians to track the migration of the DCs to the draining inguinal lymph nodes via scintigraphy. This specific formulation is being evaluated in the Phase II I-ATTAC clinical trial, led by Mustafa Khasraw, which investigates the vaccine's efficacy in newly diagnosed CMV-positive, MGMT-unmethylated glioblastoma patients. The vaccine is typically administered intradermally following pre-conditioning with tetanus-diphtheria toxoid and is often co-administered with GM-CSF as an adjuvant.
