**111In-ABD147** is a radiopharmaceutical imaging agent consisting of a high-affinity antigen binding domain targeting Delta-like ligand 3 (DLL3), fused to an engineered humanized Fc antibody and bio-conjugated with a DOTA chelator to the radioisotope Indium-111. DLL3 is expressed on the cell surface of various neuroendocrine tumors, especially small cell lung cancer (SCLC) and large cell neuroendocrine carcinoma (LCNEC), but is rarely found on nonmalignant cells[4][2][1]. This agent is designed specifically for single-photon emission computed tomography (SPECT) imaging to map DLL3-positive tumor lesions and guide the dosimetry for subsequent radiotherapeutic treatment. 111In-ABD147 binds DLL3 with high affinity (KD of 2.8 nM)[2], is rapidly cleared from normal tissues, and preferentially accumulates in DLL3-positive tumors[2][4]. Used primarily as an imaging agent in clinical development, its therapeutic counterpart, 225Ac-ABD147, utilizes the same targeting domain conjugated to Actinium-225 for radiotherapeutic intervention[1][2][3][7].
