111In-D6-B483 is a radiolabeled diabody (a non-covalent dimer of single-chain Fv fragments) derived from the human monoclonal antibody B483. It specifically targets Carbonic Anhydrase IX (CAIX), a cell-surface enzyme that is highly overexpressed in clear cell renal cell carcinoma (ccRCC) and other hypoxic tumors, while showing minimal expression in normal tissues. The diabody format (D6) is engineered for rapid blood clearance and high tumor penetration, which are advantageous for diagnostic imaging compared to full-length antibodies. Labeled with the gamma-emitting isotope Indium-111 (111In), it is utilized for Single Photon Emission Computed Tomography (SPECT) imaging to detect CAIX-positive lesions, assess tumor hypoxia, and potentially select patients for CAIX-targeted therapies.
