The 111In-DOTA-anti-CEA antibody is a radiolabeled monoclonal antibody conjugate designed for diagnostic imaging and potentially therapeutic applications in oncology. It consists of a humanized IgG1 monoclonal antibody (commonly M5A or hMN14) that targets carcinoembryonic antigen (CEA), a tumor-associated glycoprotein overexpressed in many cancers such as colorectal and pancreatic cancer. The antibody is conjugated to the chelator DOTA (1,4,7,10-tetraazacyclododecane-N,N',N'',N''' tetraacetic acid), which stably binds the radioisotope indium-111 (^111In). This allows for sensitive detection of CEA-expressing tumors using nuclear medicine imaging techniques. The mechanism involves specific binding to CEA on tumor cells followed by gamma emission from ^111In for SPECT imaging. It is being developed primarily as a diagnostic radiopharmaceutical but may also be used in pre-targeted radioimmunotherapy strategies[1][3][4][5].
