111In-DOTA-anti-CEA minibody is a radiolabeled antibody fragment used in preclinical research for tumor imaging. It consists of the T84.66 minibody, an engineered anti-carcinoembryonic antigen (CEA) fragment (scFv-CH3 dimer) with a molecular weight of approximately 80 kDa. The minibody is conjugated with the chelator DOTA (1,4,7,10-tetraazacyclododecane-N,N',N'',N'''-tetraacetic acid) and labeled with the radioisotope indium-111 (111In). This biologic imaging agent is designed to provide rapid tumor targeting and efficient blood clearance compared to intact antibodies, making it suitable for radioimmunoscintigraphy (RIS) of CEA-positive tumors. Research studies, including biodistribution and imaging in mouse xenograft models of human colorectal cancer, have been conducted at institutions such as the City of Hope National Medical Center.
