111In-DOTA-anti-CEA scFv-Fc is a radiopharmaceutical designed for the targeted imaging of tumors expressing carcinoembryonic antigen (CEA). It consists of a humanized single-chain variable fragment fused to an Fc region (scFv-Fc), creating a construct with a molecular weight of approximately 105 kDa. This format provides a balance between the rapid clearance of smaller fragments and the high tumor uptake of intact antibodies. The antibody fragment is conjugated to the chelator DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) and labeled with the gamma-emitting radionuclide Indium-111 (111In). A specific variant of this drug incorporates an I253A mutation in the CH2 domain of the Fc region, which eliminates binding to the neonatal Fc receptor (FcRn). This modification significantly accelerates blood clearance and reduces background radiation, thereby improving the tumor-to-background ratio for diagnostic imaging in conditions such as colorectal cancer.
