111In-DOTA-hTAB004 is a radiolabeled humanized monoclonal antibody being developed by OncoTab for the diagnostic imaging of tumors expressing transformed MUC1 (tMUC1). The agent consists of the hTAB004 antibody conjugated to a DOTA chelator and radiolabeled with Indium-111, enabling tumor localization via Single-Photon Emission Computed Tomography (SPECT). tMUC1 is a cancer-specific glycoform of MUC1 that is highly overexpressed in triple-negative breast cancer (TNBC) and pancreatic ductal adenocarcinoma, but absent in normal tissues. This agent is being developed as a companion diagnostic for tMUC1-targeted therapies, particularly the alpha-particle therapeutic 225Ac-DOTA-hTAB004, forming a theranostic pair for precision oncology. Preclinical studies have demonstrated high tumor accumulation and favorable biodistribution in orthotopic TNBC models.
