**111In-DTPA-NLS-CSL360** is a preclinical, Auger electron-emitting radioimmunoconjugate for **acute myeloid leukemia**. It comprises the chimeric anti-CD123 monoclonal antibody CSL360, conjugated to diethylenetriaminepentaacetic acid for chelation of indium-111 and modified with SV40-derived nuclear localization sequence peptides. The agent preferentially recognizes the CD123-positive and CD131-negative leukemia stem-cell phenotype, undergoes cellular internalization and nuclear transport, and delivers short-range Auger-electron radiation that induces lethal DNA damage. It has been evaluated in AML cell models and AML-engrafted immunodeficient mice, including studies of radioimmunotherapy and microSPECT imaging.
