Overview

111In-His6-G3 is a radiolabeled designed ankyrin repeat protein (DARPin) developed for the non-invasive molecular imaging of human epidermal growth factor receptor 2 (HER2) expression in cancers such as breast and gastric cancer. The agent consists of the G3 DARPin, which binds to a membrane-proximal epitope on HER2 with picomolar affinity, tagged with a hexahistidine (His6) sequence at the N-terminus and conjugated to the radioactive isotope indium-111 (111In) via a C-terminal DOTA chelator. Developed by researchers at University College London (UCL), Barts Cancer Institute, and the University of Zurich, 111In-His6-G3 is designed for single-photon emission computed tomography (SPECT) imaging to assess HER2 status in vivo, potentially aiding in patient stratification for HER2-targeted therapies like trastuzumab.
