111In-labeled dendritic cells are a diagnostic and therapeutic component of the ELEVATE clinical trial (NCT02366728) for newly diagnosed glioblastoma. These autologous dendritic cells are pulsed with mRNA encoding the Cytomegalovirus (CMV) pp65 antigen fused to a Lysosomal-Associated Membrane Protein (LAMP) signal and contain Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF). The cells are labeled with the radioisotope Indium-111 (111In) to allow for tracking of their migration to inguinal lymph nodes via SPECT/CT imaging. This cell-based immunotherapy aims to stimulate a T-cell mediated immune response against glioblastoma cells, which frequently express the CMV pp65 antigen. The trial specifically investigates whether pre-conditioning the injection site with tetanus-diphtheria (Td) toxoid enhances the migration and clinical efficacy of the vaccine.
