Overview

11C-YJH08 is a radiolabeled small molecule diagnostic agent used as a positron emission tomography (PET) tracer for imaging glucocorticoid receptor (GR) expression in vivo. It consists of the synthetic GR agonist YJH08 labeled with the positron-emitting isotope carbon C 11. The compound binds potently and selectively to the ligand-binding domain of GR, enabling noninvasive visualization and quantification of GR levels in tissues and tumors. This makes it useful for assessing altered GR expression and pharmacodynamics in cancer patients—particularly those with advanced malignant solid neoplasms or metastatic castration-resistant prostate cancer (mCRPC). The tracer has demonstrated high affinity for GR (Ki ~0.4 nM), rapid blood clearance, high tissue uptake including brain penetration, and specificity confirmed by competition studies. It is being developed as a companion diagnostic to guide therapy targeting the glucocorticoid receptor pathway[1][3][4][5][6][7].
