Conjugated linoleic acids (CLAs) are a group of positional and geometric isomers of linoleic acid found naturally in meat and dairy products from ruminant animals. The trans-10,cis-12 isomer (t10,c12-CLA), also referred to as 12-conjugated linoleic acid, is one of the two main biologically active forms present in commercial CLA supplements. This specific isomer has been studied for its anti-obesity effects—reducing body fat mass—and potential roles in modulating lipid metabolism and gene expression[5][6][8]. Mechanistically, t10,c12-CLA can decrease stearoyl-CoA desaturase activity (involved in fatty acid metabolism), reduce prostaglandin synthesis (notably PGE2), lower serum HDL cholesterol levels, and affect insulin-like growth factor-II secretion[6][8]. However, it may also increase risk factors for type 2 diabetes and heart disease by inducing hyperinsulinemia and liver steatosis[1][5]. While animal studies suggest possible benefits such as reduced inflammation or cancer risk modulation, human data remain mixed regarding efficacy for weight loss or other indications[1][4][8].
