12-LOX siRNA is a small interfering RNA (siRNA) designed to silence the expression of the 12-lipoxygenase (12-LOX) enzyme, specifically the human 12-HETE generating enzyme (ALOX12). In preclinical research, particularly in ovarian cancer models, the down-regulation of 12-LOX using siRNA has been shown to reduce tumor cell growth, survival, and cancer-induced angiogenesis. By inhibiting the production of 12-HETE, 12-LOX siRNA reduces the expression of vascular endothelial growth factor (VEGF), thereby impairing the angiogenic potential of cancer cells. While primarily used as a research tool to validate 12-LOX as a therapeutic target, it represents a potential RNA-based therapeutic approach for epithelial ovarian cancer and other malignancies where the lipoxygenase pathway is upregulated.
