Overview

12328zT2 is an investigational chimeric antigen receptor (CAR) T-cell therapy developed by Wellington Zhaotai, currently in preclinical development for the treatment of acute myeloid leukaemia (AML). The therapy is engineered to target CD123 (interleukin-3 receptor alpha chain), a cell surface antigen frequently overexpressed on AML blasts and leukemic stem cells compared to normal hematopoietic stem cells. The CAR construct incorporates a CD28 co-stimulatory domain and a CD3-zeta signaling domain (indicated by the '28z' designation) to promote robust T-cell activation, proliferation, and effector function upon binding to CD123-positive cells. By targeting CD123, 12328zT2 aims to eliminate the leukemic cell population while potentially sparing normal hematopoietic cells that lack high-level CD123 expression.
