123BBzT2 is a preclinical chimeric antigen receptor (CAR) T-cell therapy developed by Wellington Zhaotai for the treatment of Acute Myeloid Leukaemia (AML). The therapy involves engineering a patient's T cells to express a CAR that specifically recognizes CD123 (Interleukin-3 receptor alpha chain), a protein frequently overexpressed on AML blasts and leukemic stem cells. The 123BBzT2 construct utilizes a 4-1BB (CD137) co-stimulatory domain alongside the CD3-zeta signaling domain, a configuration intended to improve the metabolic profile, persistence, and sustained anti-tumor activity of the T cells compared to CD28-based constructs.
